Evaluation of histomorphometric changes of the seminal vesicles of young and old adult rats after receiving sesame seeds (Sesamum indicum L.)

Background & aim: Sesame seeds contain phytoestrogen and antioxidant compounds that can affect the reproductive system. Due to the importance of the seminal vesicle in male reproductive activities, the aim of this study was to determine and evaluate the histomorphometric changes of the seminal vesicles of young and old adult rats after receiving sesame seeds.
 
Methods: In the present experimental study conducted in 2017, 40 young and old rats were included in the study and divided into 4 equal groups. The young and old control groups were each divided into 2 groups (30 days and 60 days) and received 28 grams of pellets daily. The young and old treatment groups were divided into 2 groups (30 days and 60 days) and received 8.5 g of sesame seeds and 19.5 g of pellets (30% and 70% of diet) daily. At the end of the breeding period, after facilitation, sampling was performed. From the seminal vesicle by standard method of preparing tissue sections, sections with a thickness of 5-6 μm were prepared and stained by hematoxylin-eosin method. On the other hand, blood testosterone level was also measured. The collected data were analyzed using one-way ANOVA.
 
Results: Histomorphometric evaluation of seminal vesicles showed that 30- and 60-day treatment with sesame in young mice significantly increased the thickness of epithelial tissue, muscle layer thickness and total wall thickness. In 30- and 60-day-old treatments in older mice, muscle layer thickness and total wall thickness showed a significant increase. Following the consumption of sesame, testosterone increased in young mice. Also, following the consumption of sesame, no significant improvement was observed in the seminal vesicle structure of older mice compared to young mice(p <0.05).
 
Conclusion: It seems that the aging process did not cause much change in the seminal vesicle tissue, but sesame seeds were able to improve the seminal vesicle structure of young and old rats, including increasing the thickness of the epithelium, muscle layer and the entire wall.
