Cytotoxic effects and evaluation of p53gene expression toward gastric cancer AGS cells treated with Palladium nanoparticles

Background & Aims:  Gastric cancer is one of the most common cancers worldwide. Recently, Palladium nanoparticles(PdNPs), are accepted as an anti-cancer factor in studies related to cancer therapy. Therefore, the present study aimed to explore cytotoxicity effects and evaluation of p53 gene expression as a tumor suppressor gene, against gastric carcinoma AGS cells treated with PdNPs.
Methods: In this study, cancerous AGS and normal HEK293 cell lines were treated with different concentrations of PdNPs(5700, 570, 57, 5.7 µg/mL) for 48 hours. The cell viability of nanoparticles wasevaluated using MTT assay. The RNA extraction and cDNA synthesiswas done. Finally, evaluation of P53gene expressionwas analyzed via real time PCR.
Results: Thecytotoxic results revealed that PdNPscould significantly decrease cell viability in AGS cell lines at higher concentrations,compared to a control. Real time PCR results presented that the expression of P53 did notsignificantly increased compared to the control group, after 48 hours.
Conclusion: PdNPscan destroy cancerous AGS cells, compared to normal HEK293 cells,but it is not resulted from an increase in P53 gene expression. More experiments are needed to find out about cell death process due to cytotoxicity of PdNPs.
