Investigation of the Dose-Dependent Effects of Nicotine on Oxidative Stress and Liver Histopathology in Male Rats

Background & aim:The increasing use of tobacco in human societies has led to widespread diseases caused by nicotine intake. Therefore, the aim of the present study was to investigate the effect of different doses of nicotine on antioxidant function, oxidative stress, and histopathological changes in the liver in male rats.

Methods: In the present experimental laboratory study conducted in 2024, 24 male Wistar rats were randomly divided into 3 groups of 8, including control (daily intraperitoneal injection of normal saline), nicotine 0.25, and nicotine 0.5 (nicotine injection at doses of 0.25 and 0.5 intraperitoneally) for 30 days. At the end of the study period, the activities of antioxidant enzymes superoxide dismutase (SOD), glutathione peroxidase (GPX), total antioxidant capacity (TAC), and malondialdehyde (MDA) levels in liver tissue, as well as serum ALT and AST enzymes were measured. The collected data were analyzed using one-way ANOVA and Tukey's post hoc test.

Results:The results indicated that nicotine administration caused a significant increase in ALT, AST and MDA levels and a decrease in serum SOD activity (p<0.05). Also, a significant decrease in SOD and GPX enzyme activities and TAC levels and an increase in liver tissue MDA levels were reported (p<0.05). In addition, the results of liver histology changes revealed that nicotine use caused congestion and hyperemia, lymphocyte infiltration and inflammation, cell degeneration, and necrosis.

Conclusion: The present study exposed that nicotine administration, through the induction of oxidative stress, lead to an increase in serum and liver tissue malondialdehyde levels and caused tissue changes and liver damage by reducing the activity of antioxidant enzymes.
