Fasciola coproantigens detection, using polyclonal antibodies produced against Fasciola hepatica and Fasciola gigantica excretory-secretory antigens

Background: Parasitological tests have limitations for diagnosing fascioliasis such as intermittent egg shedding, long prepatent, and patent time. Serological tests based on antibody detection do not differentiate between current and past infection. Detection of Fasciola coproantigen, rather than the other circulatory antigens, seems to be a reliable approach in diagnosing fascioliasis. Considering the points mentioned above, the current study aimed to develop an appropriate antigen detection assay (sandwich ELISA), using polyclonal antibodies raised against excretory-secretory (ES) antigens of Fasciola species for diagnosis of fascioliasis. 
Materials and methods: Fasciola worms were collected from the liver of infected animals. Fasciola species were determined using RFLP-PCR. ES antigens were prepared. Two rabbits for each antigen were immunized. Also, rabbit IgG antibodies raised against Fasciola ES antigens were purified using affinity chromatography. Then, polyclonal antibodies were labeled with Horseradish peroxidase. Coproantigens were prepared from 99 stool samples of ruminants, including Fasciola infected stools samples (n= 30), stool samples with other parasites (n= 39), and negative control stool samples (n= 30) of healthy ruminants. In this study for the detection of Fasciola coproantigen, two systems of sandwich ELISA were developed.
Results: Sandwich ELISA using polyclonal antibodies raised against Fasciola hepatica (F.hepatica) and Fasciola gigantica (F.gigantica) ES antigens for diagnosis of Fasciola coproantigens showed sensitivity and specificity 96.67%, 89.86 and 96.67%, 91.3%, respectively. Two produced polyclonal antibodies showed good agreement (ҡ > 0.9) for the diagnosis of fascioliasis.
Conclusion: Findings of this study determined that the produced polyclonal antibodies raised against F. hepatica and F. gigantica ES antigens have appropriate performance for the detection of Fasciola coproantigens and can be used in the ELISA system for diagnosis of fascioliasis
