Simultaneous effect of aerobic training and octopamine on tissue levels of NLRP1 inflammasome, leptin, carbonyl protonil and histology of brown adipose tissue changes in rats fed with deep fried oil

Background and Objective: Consumption of crunchy foods and lack of nutritional hygiene causes destruction of adipose tissue homeostasis and secretion of inflammatory factors. Aerobic exercise is one of the ways to control inflammation adipose tissue. So,the aim of this study was to investigation the simultaneous effect of aerobic exercise and Octopamine on NLR Family Pyrin Domain Containing 1 (NLRP1), leptin, carbonyl protonil of brown adipose tissue changes in rats fed with deep fried oil.
Materials and Methodsforty male Wistar rat (age 20 weeks; weight 350 ±20 g) randomly in devider into healthy control groups (n = 8), poisoned control with DFO (n = 8), poisoned + aerobic exercise (8 = n), poisoned + octapamine (8 = n) and poison + aerobic exercise + octopamine (8 = n). The DFO was orally fed to the relevant rats for 4 weeks. The octopamine in dose of 81 mg / kg dissolved in 9% normal salineused and intraperitoneal (IP) injected. Aerobic training groups also exercise on a treadmill for 4 weeks with increasing intensity in the first week of 50% aerobic exercise and in the last week the intensity of training reached 65% of aerobic strength. The concentration of leptin, protonyl carbonyl concentrations was investigated using ELISA method and changes in brown adipose tissue using hematoxylin-eosin and IHC staining methods. Real time PCR and immunohistochemistry methods were used to determine changes in the NLRP1 gene and protein, respectively. For data analysis, two-way analysis of variance was used with a significance of P ≤0.05.
Results: In all toxic groups seen a significant increase in the expression of NLRP1 gene and protein, as well as an increase in carbonyl and liptin values compared to the healthy control group (p≤0.05). The results showed that the expression of NLRP1 gene and protein in the DFO-aerobic and DFO-octapamine groups decreased significantly compared to the DFO sole group (p≤0.05). Also, the expression of NLRP1 protein in the DFO+aerobic+octapamine group was significantly different compared to the DFO control group (p≤0.05), while the expression of this gene in the DFO + aerobic + octapamine group did not decrease significantly compared to the toxic group. The results showed a significant difference in the concentration of carbonyl protein in the DFO-aerobic and DFO-octapamine groups compared to the DFO sole group, but this result was not observed in the DFO-aerobic exercise + octapamine. The concentration of leptin in DFO- groups (poisoning + octapamine and poisoning + aerobic exercise + octapamine) had a significant decrease compared to the DFO sole group.
Conclusion: It seems that, using octopamine supplementation with aerobic exercise, by controlling NLRP1 and decreasing leptin and carbonyl, may be effective in improve adipose tissue risk factors.
