In vitro and in vivo study of the effects of Lawsonia inermis on Toxoplasma gondii

Background: Toxoplasmosis is a common parasitic disease between humans and animals that is caused by the protozoan Toxoplasma gondii. The Henna plant (Lowsonia inermis) is effective against a wide range of diseases. The first-line drugs for treatment of toxoplasmosis are pyrimethamine and sulfadiazine that have side effects. On the other hand, despite the increasing resistance of parasites to chemical drugs, the use of medicinal herbs and traditional treatments has become widespread. This study undertaken to evaluate in vivo and in vitro effect of Lawsonia inermis on Toxoplasma gondii.
Methods: The henna leaves were prepared and extracted using 80% ethanol by percolation method. Antitoxoplasma effects of Lawsonia inermis extracts (2, 4, 8, 16, 32, 64, 128, 256, 512 µg/ml) on Toxoplasma tachyzoites were evaluated using Propidium Iodide staining and flow cytometry method. To analyze the in vivo anti-toxoplasma activity, 35 Balb/C mice were infected with tachyzoites of Toxoplasma intradermally. Different concentrations of Lawsonia inermis extract (16, 32, 64,128 mg/kg) were given to four the treated groups of mice. One group receiving sulfadiazine as a positive control and one group without prescribing as a negative control were considered.
Results: IC50 value of Lawsonia inermis extract was 100 µg/ml in vitro. There was no significant difference in the mean survival time of the mice in the treated groups and control group (P≥0.05). Therefore,   Lawsonia inermis extracts did not show any activity in vivo.
Conclusion: The results of this study showed that the effect of Henna hydroalcoholic extract is not significant in vivo, although it is effective in vitro. It seems that kind of administration and matabolization of the compounds in the extract causes the different between in vitro and in vivo results.
