The preventive effect of one course of resistance training with injections of vitamin D3 on the levels of klotho in the brain tissue of female rats with experimental autoimmune encephalomyelitis

Background & objectives: This study aimed to determine the pretreatment effect of 6-week Resistance exercise with injections of vitamin D3 supplementation on the levels of klotho in female Lewis rats with experimental autoimmune encephalomyelitis. Methods: In this experimental study that conducted on female Lewis rats, animals were randomly divided into 9 groups (n=5 per group), including, healthy control, EAE control, sham, Resistance training, EAE Resistance training, vitamin D3, EAE vitamin D3, combination, EAE combination. Resistance training program included climbing a ladder with tail-loaded weights. In addition, vitamin D3 groups received vitamin D3 supplementation (2ng/0.2mg sesame oil/every two days) intraperitoneal for 2 weeks. To induce EAE model, to each animal in 4.0 ml of solution guinea pig spinal cord and equally adjuvant and, was injected klotho levels were measured by ELISA. Analyze data using ONE WAY ANOVA and Tukey test was performed with a significance level of p≤0/05. Results: The results of this study showed that Mean serum levels of klotho in the brain tissue of control group were significantly different from control group(p=0/001).  Klotho levels in the resistance training group was significantly higher than the control group (p=0/001). also there was a significant increase in klotho in supplementation and combination patient group compared to the control group (Respectively P=0/001؛ P=0/006). Also the difference between the training, supplementation and combination groups was not significant (p≥0/05). Conclusion: The result of this study show that pretreatment with the Resistance training and injection of vitamin D3, alone and in combined together was able to prevent the reduction of klotho's levels against EAE induction. As a result, this protocol is likely to have a protective effect against MS.
