Correlation between MicroRNA129-2 expression and cervical cancer patients infected with HPV

Abstract

Introduction: Cervical cancer is the fourth leading worldwide cancer and the first cause of mortality among women and gets seventh rank among all the types of cancers. Human Papilloma Virus (HPV) infection is one of the risk factors leading to cervical cancer. MicroRNAs have been suggested as new marker for cervical cancer diagnosis. MicroRNA129-2 is a cell proliferation and migration suppresser. The aim of this study was to determine the expression of MicroRNA129-2 in infected cervical cancer patients with women with no HPV infection and compare them with normal healthy woman as control.

Methods: This present study was composed of 20 cancerous paraffin-embedded tissues from HPV infected cervical cancer patients, 20 cervical cancer with not infection and 10 normal Pap smear were collected from Mirzakochak Khan hospital, Tehran in 1394. RNA was extracted and miR129-2 expression was investigated using Real time PCR among groups.

Result: MicroRNA129-2 expression was decreased in infected cancerous tissues compared with patients with no infection (P=0.0002). Also, we found a statistically significant reduction in MicroRNA129-2 expression in cervical cancer patients compared with healthy group (P >0.001). Analysis was performed using Graph pad prism 6 software and no significant association was observed between MicroRNA129-2 expression with age (P=0.93) and stage of disease (P=0.386).

Conclusion: According to the results it can be concluded that MicroRNA129-2 expression was strongly decreased in tumor samples. So determination of miR129-2 expression may offer valuable diagnosis information for early detection and prognoses of patients with cervical cancer.
