Antiprolifration effect of silver nanoparticles syntethised using Centlla asiatica on breast cancer cell line

Background and Objectives: The use of plants for the synthesis of nanoparticles, because of not using pollutants and toxins, is considered an eco-friendly method. The purpose of this study is silver nanoparticles biosynthesis using centella asiatica extract and evaluating the toxicity of synthesized silver nanoparticles on breast cancer cell line.
Materials and Methods: For synthesizing silver nanoparticles, the leaf extract of Centella asiatica was used. Phytomorphology of bio silver nanoparticles using TEM (Transmission Electron Microscopy) and SEM (Scanning Electron Microscope) analysis was evaluated. Antiplolifration effects of bio nanoparticles in breast cancer (MCF-7) and normal cell (HEK293) were evaluated in 24 hours by MTT assay. Annexin V kit was used for apoptosis test by flocytometry. Based on two way ANOVA, the control groups were compared with the control group.  
Results:The average size of nanoparticles are 19.17 nm. The IC50 value for silver nanoparticles was 8.76 µg/ml during 24 h on breast cell line, which was significant comparing to the control group. Increasing apoptosis was observed in cells treated with biological nanoparticles at IC50 concentration compared to control cells (P <0.001). Also, induction of apoptosis in MCF-7 cells was more than necrotizing compared to control (P <0.001).
Conclusion In this study, for the first time, we investigated the effect of the silver nanoparticles synthesized from C. asiatica extract on breast cancer cell line and results suggest that synthesized silver nanoparticles have cytotoxicity and apoptosis induction effects on breast cancer cells that depends on time and dosage pattern.
