Investigating the effect of cytotoxicity of probiotic bacteria isolated from bee honey stomach on HT29 clone cancer cell line and analyzing the expression of apoptotic genes Bcl2, BAX and P53

Introduction: Colorectal cancer is a global health burden that requires the discovery of new treatment approaches. Probiotic bacteria, especially those isolated from honey bee stomachs, have attracted attention for their potential anticancer properties. This study was managed with the aim of investigating the cytotoxic effect of probiotic bacteria isolated from the stomach of bee honey on the cancer cell line clone HT-29 and variation the expression of key apoptosis genes Bcl2, BAX and P53.
Methods: Probiotic bacteria were isolated from bee stomachs. Identification of bacteria was evaluated by molecular and biochemical techniques. The cytotoxic effect of these bacteria on HT-29 cells was measured by using MTT assay. Real-time PCR method was used to evaluate the expression of Bcl2, BAX and P53 genes. Data was analyzed Data was statistical analyzed.
Results: Among the 30 isolated colonies from probiotic strains, were identified Lactobacillus plantarum and Lactobacillus acidophilus that significantly decreased the viability of HT-29 cells compared to the control. Lactobacillus plantarum, there was a significant decrease in the expression of the anti-apoptotic Bcl-2 gene and an increase in the expression of the pro-apoptotic BAX and P53 genes in the treated cells (P<0.001).
Discussion: Lactobacillus plantarum strains isolated from honey bee stomachs showed strong anti-proliferative effects on colorectal cancer cells through changing the expression of apoptotic genes. These probiotic strains could have potential therapeutic agents against colon cancer. Further studies for this matter were recommended.
