Protective effect of Bone marrow mesenchymal cells on Testis Histologic and Histomorphometric properties in Mice following Spermatic cord Torsion-Detorsion

Background and aim: Regarding that, the disturbance in testicular blood circulation and consequently testis ischemia can disrupt fertility. In this study, the protective effect of bone marrow mesenchymal cells on the testicular histomorphometry following spermatic cord Torsion-Detorsion was investigated comparatively
Methods: In this experimental study, 30 adult mice were randomly divided into three groups. In Con-sham group, the laparotomy has been done without the torsion-detorsion (T-D). T-D group, in this group torsion-detorsion was created in the left testicular cord and after repefusion, 10µl PBS injected into the rete testis. In (T-D+Msc) group, after left testicular cord torsion-detorsion mesenchymal cells (5×105 in 10µL) was injected into rete-testis. At the end of the treatment period, (35 days) the left testis were fixed and paraffin sections stained with H&E and Masson-trichrom for the number of blood vessels. The different histomorphometric parameters were calculated. Obtained data were analyzed by SPSS software and ANOVA statistic method and Tukey post hoc test.( P<0.05).
 Results: This study showed that the different histomorphometric parameters such as spermatogenesis indices, mean number of active Sertoli and Lydig cells, seminiferous tubules diameter, thickness of germinal epithelium and testicular capsule, in the treatment group (T-D+Msc), significantly increased in comparison to the T-D group (P<0.05). The examination of the mean number of vessels in testis, showed that in the T-D group was seen significant decrease compared to the (T-D+Msc) group (P<0.05).
Conclusions: Histomorphometrical studies have shown that the use of mesenchymal cells in the process of improvement of the impact due to testicular cord torsion-detorsion can be effective by increasing angiogenesis and tissue repairing.
