Trigonella foenum-graecum) on changes in sex hormones in rat model of menopause فیزیولوژی Physiology پژوهشي Research رزمینه و <a>هدف</a> <div> <hr align="left" class="msocomoff" size="1" width="33%" > <div> <div class="msocomtxt" id="_com_1" language="JavaScript" on<illegal tag>mouseout="msoCommentHide('_com_1')" on<illegal tag>mouseover="msoCommentShow('_anchor_1','_com_1')"><a name="_msocom_1"></a> <a class="msocomoff" href="#_msoanchor_1">[ma1]</a>محدودیزمینه و <a>هدف</a><a class="msocomanchor" href="#_msocom_1" id="_anchor_1" language="JavaScript" name="_msoanchor_1" on<illegal tag>mouseout="msoCommentHide('_com_1')" on<illegal tag>mouseover="msoCommentShow('_anchor_1','_com_1')">[ma1]</a> : یائسگی همراه با کاهش عملکرد تخمدان و نقصان هورمونهای جنسی است که کمابیش بر سلامت جسم و روح زنان تأثیر می‌گذارد. ارائه راهکارهای درمانی با کمترین عوارض جانبی حائز اهمیت است و گیاه شنبلیله (Trigonella foenum- graecum) بدلیل داشتن ترکیبات فیتواستروژنی و برخی ترکیبات ضداکسیدانی، شاید گزینه‌ مناسبی باشد. روش بررسی: پس از تهیه <a>عصاره</a><a class="msocomanchor" href="#_msocom_2" id="_anchor_2" language="JavaScript" name="_msoanchor_2" on<illegal tag>mouseout="msoCommentHide('_com_2')" on<illegal tag>mouseover="msoCommentShow('_anchor_2','_com_2')">[ma2]</a>  هیدروالکلی برگ شنبلیله و تعیین دوز کشنده، 48 سر موش ‌صحرایی ماده بالغ با وزن 200-180 گرم انتخاب و در 6 گروه&lrm; شامل <a>کنترل</a><a class="msocomanchor" href="#_msocom_3" id="_anchor_3" language="JavaScript" name="_msoanchor_3" on<illegal tag>mouseout="msoCommentHide('_com_3')" on<illegal tag>mouseover="msoCommentShow('_anchor_3','_com_3')">[ma3]</a> ، یائسه بدون تیمار و 2 گروه تیمار شامل موش&lrm;های مدل یائسه و دو گروه تیمار غیریائسه دسته بندی شدند. القای یائسگی با تزریق روزانه  mg/kg.bw 7 از داروی سیس&lrm;پلاتین طی 5 روز انجام شد و تیمار موش&lrm;ها در هر دو مدل با دوزهای mg/kg.bw (250 و500) از عصاره هیدروالکلی شنبلیله <a class="msocomanchor" href="#_msocom_4" id="_anchor_4" language="JavaScript" name="_msoanchor_4" on<illegal tag>mouseout="msoCommentHide('_com_4')" on<illegal tag>mouseover="msoCommentShow('_anchor_4','_com_4')">[ma4]</a> بصورت گاواژ روزانه انجام شد. در پایان دوره تیمار(24 روز) و پس از خونگیری از قلب موش&lrm;‌ها، غلظت سرمی گنادوتروپین&lrm;ها و هورمون&lrm;های جنسی با روش رادیوایمونواسی و برخی فاکتورهای استرس اکسیداتیو با روش الیزا اندازه&lrm;‌گیری شد. داده&lrm;‌های حاصل با نرم‌افزار SPSS و آزمون ANOVA تحلیل و با آزمون توکی در سطح معناداری 0.05≤Pمقایسه شد. یافته&lrm;ها: میزان هورمون&lrm;های تستسترون، پروژسترون و استرادیول در سرم موش&lrm;های مدل یائسه در مقایسه با گروه کنترل، کاهش معنادار و هورمون لوتئینه&lrm; کننده و هورمون محرکه &lrm;فولیکول افزایش معنی&lrm;دار (P≤0.05) نشان دادند که با مصرف عصاره هیدروالکلی برگ شنبلیله (بخصوص در دوز mg/kg.bw500) در سطح معناداری تعدیل شد. افزایش آنزیم مالون&lrm;‌دی&lrm;‌آلدهید در گروه‌های تحت القای یائسگی و جبران آن با مصرف عصاره مذکور و همچنین افزایش ظرفیت آنتی&lrm;‌اکسیدانی&lrm;‌تام و سوپراکسید دیسموتاز بخصوص در موش&lrm;های غیریائسه در مقایسه با گروه کنترل مشاهده گردید. <a>نتیجه‌</a><a class="msocomanchor" href="#_msocom_1" id="_anchor_1" language="JavaScript" name="_msoanchor_1" on<illegal tag>mouseout="msoCommentHide('_com_1')" on<illegal tag>mouseover="msoCommentShow('_anchor_1','_com_1')">[ma1]</a> &lrm;گیری: با توجه به اثر تعدیلی عصاره هیدروالکلی برگ شنبلیله بر هورمون&lrm;های مورد بررسی بخصوص در موش&lrm;های غیریائسه و کاهش مالون&lrm; دی&lrm;آلدهید و افزایش آنزیم&lrm;های آنتی &lrm;اکسیدانی در نتیجه مصرف عصاره مذکور حتی در گروه&lrm;های یائسه، میتوان به اثر مثیت این گیاه در تعدیل هورمون‌های زنانه و کاهش عوارض نامطلوب یائسگی با تأکید بر خواص آنتی&lrm; اکسیدانی تا حدودی امیدوار بود. <div> <hr align="left" class="msocomoff" size="1" width="33%" > <div> <div class="msocomtxt" id="_com_1" language="JavaScript" on<illegal tag>mouseout="msoCommentHide('_com_1')" on<illegal tag>mouseover="msoCommentShow('_anchor_1','_com_1')"><a name="_msocom_1"></a> <a class="msocomoff" href="#_msoanchor_1">[ma1]</a>اصلا</div> </div> </div> <a>نتیجه‌</a><a class="msocomanchor" href="#_msocom_1" id="_anchor_1" language="JavaScript" name="_msoanchor_1" on<illegal tag>mouseout="msoCommentHide('_com_1')" on<illegal tag>mouseover="msoCommentShow('_anchor_1','_com_1')">[ma1]</a>  <div class="msocomtxt" id="_com_1" language="JavaScript" on<illegal tag>mouseout="msoCommentHide('_com_1')" on<illegal tag>mouseover="msoCommentShow('_anchor_1','_com_1')"><a name="_msocom_1"></a> <a class="msocomoff" href="#_msoanchor_1">[ma1]</a>م</div> </div> </div> </div> <div dir="ltr" style="text-align: right;"></div> Background and Objective: Menopause is accompanied by ovarian dysfunction and a decrease in sex hormones, which more or less affects the physical and mental health of women. It is important to provide treatment solutions with minimal side effects. Fenugreek (Trigonella foenum-graecum) may be a good option, because it contains phytoestrogenic and some antioxidant compounds. Methods: After preparing hydroalcoholic extract of fenugreek leaves and determining the lethal dose, 48 adult female desert rats weighing 180-200 grams were selected and divided into 6 groups include control, untreated menopausal rats, and 2 treatment groups include menopausal model rats and two non-menopausal treatment groups. Menopause induction was performed by daily injection of 7mg/kg.bw of cisplatin for 5 days, and rats in both models were treated with doses of 250 and 500mg/kg.bw of fenugreek hydroalcoholic extract by daily gavage. At the end of the treatment period (24 days) and after taking blood from rat’s hearts, the serum concentration of gonadotropins and sex hormones was measured by radioimmunoassay method, and some oxidative stress factors were measured by ELISA method. The obtained data were analyzed with SPSS software and ANOVA test and compared with Tukey's test at a significance level of P≤0.05. Results: Testosterone, progesterone, and estradiol hormones were significantly decreased in menopausal model rats compared to control, but luteinizing hormone and follicle stimulating hormone showed a significant increase, which was significantly modulated by the consumption of fenugreek leaf hydroalcoholic extract (especially at a dose of 500 mg/kg.bw). An increase in malondialdehyde enzyme was observed in menopause induced rats, and its compensation with the consumption of the mentioned extract, as well as an increase in total antioxidant capacity and superoxide dismutase, especially in non-menopausal rats compared to the control group. Conclusion: Considering the modulating effect of the hydroalcoholic extract of fenugreek leaf on the examined hormones, especially in non-menopausal rats, and the reduction of malondialdehyde and the increase of antioxidant enzymes as a result of the consumption of the said extract even in menopausal groups, we can hope for the effect of this plant in reducing the adverse effects of menopause by emphasizing its antioxidant properties to some extent.Background and Objective: Menopause is accompanied by ovarian dysfunction and a decrease in sex hormones, which more or less affects the physical and mental health of women.
