fa تأثیر نانوپلیمر قالب مولکولی ـ نانوذرات اکسید روی(Zn) آغشته شده به اسانس مرزه خوزستانی بر میزان بیان ژن lasR در جدایه‌های بالینی پسودوموناس آئروژینوزا: یک مطالعه آزمایشگاهی The Effect of Molecular Template Nanopolymer-zinc oxide(Zn) Nanoparticles Impregnated with Khuzestani Essential Oil on lasR Gene Expression in Clinical Isolates of Pseudomonas aeruginosa: a laboratory study میکروبیولوژی Microbiology پژوهشي Research زمینه و هدف: در این مطالعه، به عنوان یک زمینه پژوهشی، بررسی کاربرد نانوذرات و گیاهان دارویی در درمان بیماری‌های ناشی از پسودموناس آئیروژینوزا مورد بررسی قرار گرفت. هدف از این مطالعه تعیین و تأثیر نانوذره روی(Zn) آغشته شده به اسانس مرزه خوزستانی((Satureja khuzestanica بر میزان بیان ژن lasR در جدایه‌های بالینی پسودوموناس آئروژینوزا تشکیل دهنده بیوفیلم در شرایط برون تنی بود. روش بررسی: این یک مطالعه تجربی می‌باشد که در سال 1399 بر روی 150 نمونه بالینی مشکوک به پسودموناس آئروژینوزا غیر تکراری و متناوب که از نمونه‌های پوست، خون و ادرار بیماران بستری در بخش‌های مختلف بیمارستان امام سجاد (ع) شهر یاسوج جمع‌آوری شد، انجام گرفت. پس از ورود 4 سویه بالینی به همراه یک سویه استاندارد پسودوموناس آئروژینوزا   (ATCC 25922)به عنوان نمونه، اسانس مرزه‌خوزستانی از شرکت دارویی باریج اسانس(کاشان، ایران) تهیه شد. سپس، پس از تعیین هویت نمونه‌ پسودوموناس آئروژینوزا جدا شده از بیماران،  PCRجهت تأیید حضور ژنlasR  و ژن DNA gyrase A ، تعیین الگوی حساسیت آنتی‌بیوتیکی، آزمون میکرودایلوشن براث، رقیق‌سازی چکربورد، آزمون آگار برای تعیین حداقل غلظت کشندگی، بررسی بیان ژنlasR  با Real time PCR، در شرایط آزمایشگاهی مورد ارزیابی قرار گرفتند. داده‌های جمع‌آوری شده بر استفاه از آزمون‌های آماری من ویتنی، تی و تی جفت نمونه‌ای تجزیه و تحلیل شدند. یافته‌ها: با توجه به نتایج الگوی حساسیت آنتی‌بیوتیکی، بیشترین مقاومت مربوط به آمیکاسین(30 میکروگرم)، کولیسین(10 میکروگرم) و سیپروفلوکسازین(5 میکروگرم) بود و کمترین مقاومت مربوط به سفتازیدین(30 میکروگرم)، و ایمی پنم(10 میکروگرم)، بود. هم‌چنین، اسانس مرزه‌خوزستانی و نانوذرات اکسید روی در برابر سویه استاندارد و جدایه‌های بالینی، دارای حداقل غلظت مهاری(256ـ512 میکروگرم/میلی لیتر) بودند. حد شاخص غلظت مهاری نسبی(FIC) برابر عدد 1 شد که نماینگر اثر افزایش است. هم‌چنین، حداقل غلظت باکتری‌کشی(MBC) چهار برابر حداقل غلظت مهاری(MIC) بود. بالاترین MIC، مربوط به نانوذرات اکسید روی و کمترین آن مربوط به استفاده هم‌زمان اسانس و نانوذره شد. در حالت ترکیبی تیمارها، به نصف غلظت MIC تیمارهای مجزا، نیاز بود. هم‌چنین، با کاهش اثر مهار کنندگی تیمارهای مورد مطالعه، تولید بیوفیلم افزایش یافت. میزان بیان ژن lasR در غلظت MIC انواع تیمارهای مطالعه متفاوت بود؛ کم‌ترین میزان بیان ژن lasR در غلظت MIC به هر یک از تیمارها به تنهایی و بیشترین به استفاده همزمان تیمارها اختصاص داشت. نتیجه‌گیری: نتایج نشان داد که سویه‌های بالینی به برخی از آنتی‌بیوتیک‌ها بسیار مقاوم بودند، اما به برخی دیگر حساس بودند. هم‌چنین، ترکیبات اسانس گیاه مرزه خوزستانی شناسایی و تجزیه شدند. اصلی‌ترین ترکیب این اسانس کارواکرول بود که خاصیت ضدعفونی کننده دارد. با استفاده از FT-IR، گروه‌های عاملی پلیمر قالب مولکولی و اسانس شناسایی شدند. نتایج نشان داد که ترکیب اسانس و نانوپلیمر روی به‌صورت جداگانه منجر به کاهش بیان ژن lasR در سویه‌های بالینی می‌شوند. با ترکیب این دو تیمار، تأثیر کاهش بیان ژن lasR قوی‌تر و اثربخش‌تر می‌شود. ‌   Background & aim: In the present study, as a research background, the application of nanoparticles and herbal medicines in the treatment of diseases caused by Pseudomonas aeruginosa was investigated. The aim of this study was to determine the effect of zinc nanoparticles (Zn) impregnated with Khuzestan savory (Satureja khuzestanica) essential oil on the expression of lasR gene in clinical isolates of Pseudomonas aeruginosa, which are biofilm-forming bacteria, under extracellular conditions. Methods: The present experimental study was conducted in 2019 on 150 non-repetitive and intermittent clinical samples suspected of Pseudomonas aeruginosa, which were collected from skin, blood, and urine samples of hospitalized patients in various wards of Imam Sajjad (AS) Hospital in Yasuj city. After the entry of 4 clinical strains along with a standard strain of Pseudomonas aeruginosa (ATCC 25922) as a sample, Khuzestan savory essential oil was prepared from Barij Essence Pharmaceutical Company (Kashan, Iran). Then, after identifying the Pseudomonas aeruginosa sample separated from the patients, PCR was performed to confirm the presence of lasR and DNA gyrase A genes, determine the antibiotic sensitivity pattern, broth microdilution test, checkerboard dilution, and agar test to determine the minimum inhibitory concentration, and real-time PCR to investigate the expression of lasR gene under laboratory conditions. The collected data were analyzed using Mann-Whitney U, T-Test, and Paired T-Test statistical tests. Results: According to the results of the antibiotic sensitivity pattern, the highest resistance was related to amikacin (30 μg), colistin (10 μg), and ciprofloxacin (5 μg), and the lowest resistance was related to ceftazidime (30 μg) and imipenem (10 μg). Also, Khuzestan savory essential oil and zinc oxide nanoparticles had a minimum inhibitory concentration of 256-512 μg/mL against standard strains and clinical isolates. The relative minimum inhibitory concentration index (FIC) was equal to 1, which represents an increase in effect. Also, the minimum bactericidal concentration (MBC) was four times the minimum inhibitory concentration (MIC). The highest MIC was related to zinc oxide nanoparticles and the lowest was related to the simultaneous use of essential oil and nanoparticles. In the combined mode of treatments, half the MIC of separate treatments was required. Also, with the reduction of the effect m. Conclusion: The results indicated that clinical strains were highly resistant to some antibiotics but sensitive to others. Also, Khuzestan savory essential oil and zinc oxide nanoparticles were effective against standard strains and clinical isolates. The main component of this essential oil was carvacrol, which has antimicrobial properties. The results showed that the combination of essential oil and nanoparticles led to a stronger and more effective reduction in lasR gene expression.
