fa بررسی بیان ژن‌هایSMAD3 وSMAD4 در بیماران مبتلا به CML و رده سلولی K562 Investigation of SMAD3 and SMAD4 Genes Expression in CML Patients and K562 Cell Line تخصصي Special پژوهشي Research زمینه و هدف: لوسمی میلوییدی مزمن یا CML یک اختلال میلوپرولیفراتیو کلونال با شاخص سیتوژنتیکی کروموزوم غیر طبیعی فیلادلفیا می‌باشد. یکی از مسیرهای پیام رسانی مهم در روند تکثیر و آپوپتوز سلول‌های سرطانی و پاتوژنز بیماری β-TGF می‌باشد که عدم تنظیم بیان اجزای آن مانند‌ خانواده SMADها نقش به سزایی در تنظیم رشد تومور و توسعه سرطان ایفا می‌کند. هدف از این مطالعه تعیین و بررسی بیان ژن‌های SMAD3  وSMAD4 در بیماران مبتلا به CML و رده سلولی K562 بود. روش بررسی:  این مطالعه توصیفی به صورت مورد ـ شاهدی می‌باشد که در سال 1400 در دانشگاه آزاد اسلامی واحد مرودشت انجام شد. جامعه آماری پژوهش شامل 80 نفر مراجعه کننده به پیوند می‌باشند که در دو گروه ؛ 40 بیمار مبتلا به CML به عنوان گروه مورد و 40 فرد سالم به عنوان کنترل که از نظر سن و جنس سازگار بودند و رده سلولی K562  انتخاب شدند. میزان بیان ژن‌های SMAD3 و SMAD4به وسیله تکنیک Real Time PCR  و سنتز cDNA  سنجیده شد و داده‌های جمع‌آوری شده با استفاده از آزمون‌های آماری T test آنالیز واریانس یک طرفه تجزیه و تحلیل شدند. یافته‌ها: ژن SMAD3دربیماران مبتلا به CML  به طور معنا داری دچار کاهش بیان بود و این نتیجه در رده سلولی K562  نیز مشاهده گردید(p<0.0001) و میزان بیان ژن SMAD4 در بیماران و رده سلولی K562  هیچگونه ارتباط معناداری با بیماری را نشان نداد(p<0.001)  مقادیر P کمتر از 0.05 در نظر گرفته شد). نتیجه‌گیری: بر طبق مطالعه انجام شده بیان برخی ژن‌ها در مسیرهای پیام رسانی می‌تواند نقش مؤثری در توسعه بیماری یا مقاومت دارویی داشته باشد. کاهش بیان SMAD3 در CML می‌تواند نشان دهنده ارتباط بیماری و اختلال در بیان ژن‌ها باشد.   Background & aim: Chronic myeloid leukemia or CML is a clonal myeloproliferative disorder with abnormal Philadelphia chromosome cytogenetic index. One of the important signaling pathways in the process of proliferation and apoptosis of cancer cells and disease pathogenesis is β-TGF, the dysregulation of the expression of its components, such as the SMAD family, plays a significant role in regulating tumor growth and cancer development. Therefore, the aim of the present study was to determine and investigate the expression of SMAD3 and SMAD4 genes in patients with CML and K562 cell line. Methods: the present descriptive case-control study was conducted in 2020 at the Islamic Azad University, Marvdasht Branch. The statistical population of the research included 80 people who referred to the link and were divided into two groups; 40 patients with CML were selected as the case group and 40 healthy individuals as controls, matched in terms of age and sex, and K562 cell line. The expression levels of SMAD3 and SMAD4 genes were measured by Real Time PCR and cDNA synthesis, and the collected data were analyzed using one-way ANOVA T-test. Results: The SMAD3 gene was significantly decreased in CML patients and this was as well observed in the K562 cell line (p<0.0001) and the level of SMAD4 gene expression in the patients and the K562 cell line did not display any significant relationship with the disease (p<0.0001). 0.001) P values less than 0.05 were considered. Conclusion: According to the results of the present study, the expression of some genes in the signaling pathways can play an effective role in the development of disease or drug resistance.
