fa تأثیر استرادیول بر شاخص استرس اکسیداتیو و سطوح نیتریک اکساید در انسداد یک طرفه میزنای در رت تخمدان برداری شده The Effect of Estradiol on Oxidative Stress Index and Nitric Oxide Levels in Unilateral Ureteral Obstruction in Ovariectomized Rats فیزیولوژی Physiology پژوهشي Research زمینه و هدف: انسداد یک‌طرفه میزنای(UUO) یک اختلال شایع در سیستم کلیوی است که ممکن است باعث آسیب غیر قابل بازگشت کلیه شود و وابسته به طول دوره انسداد و میزان آسیب بافت کلیه و اختلال در عملکرد آن می‌باشد. هدف از این مطالعه تعیین و تأثیر استرادیول بر شاخص استرس اکسیداتیو و سطوح نیتریک اکساید در انسداد یک طرفه میزنای در رت تخمدان برداری شده بود.   روش بررسی: در این مطالعه تجربی که در سال 1396 انجام شد، 84 سر رت ویستار اوورکتومی شده(20±180 گرم) به صورت تصادفی به 14 گروه 6 تایی تقسیم شدند. گروه 1(گروه شم) بدون جراحی بود و تمام گروه‌های 2 تا 14 دچار انسداد حالب یک طرفه شده و بعضی از آنها تحت درمان دارو قرار گرفتند. بدین ترتیب که؛ گروه 4-2، 3 روز پس از UUO و دریافت دارو(روغن کنجد، 1/0 و 5/0 میلی‌گرم بر کیلوگرم استرادیول) به ترتیب ساکریفایز شدند، گروه 7ـ5، 3 روز پس از UUO و دریافت دارو رفع انسداد شده(RUUO) و روز بعد ساکریفایز شدند، گروه‌های 9ـ8 و 12ـ10 به ترتیب یک و 3 روز بعد از RUUO و دریافت استرادیول ساکریفایز شدند و گروه‌های 14ـ13، 3 روز بعد از UUO و RUUO و دریافت دارو در هر دو مدل ساکریفایز شدند. آزمایشات تشخیص کمی متابولیت پایدار نیتریک اکساید و مالون دی‌آلدهید در نمونه خون و بافت به وسیله کیت‌های اختصاصی صورت گرفت. داده‌های جمع‌آوری شده با استفاده از آزمون‌های آماری و تست ال اس دی تجزیه و تحلیل شدند.   یافته‌ها: نتایج مطالعه حاضر نشان داد که در مدل UUO، سطح نیتریت و MDA  بافت کلیه در گروه‌های انسدادی نسبت به گروه شم به صورت معنی‌داری کاهش یافت( 05/0p<). با این حال تجویز دوزهای مختلف استرادیول تغییری در سطح سرمی و بافتی نیتریت و MDA بین گروه‌های درمانی و کنترل در مدل  UUOو RUUO ایجاد نکرد( 05/0p>).   نتیجه‌گیری: با توجه به نتایج به دست آمده می‌توان بیان کرد که تجویز استرادیول در زمان‌های UUO و RUUO  اثری برسطوح نیتریت و MDA نداشت. اگرچه احتمالاً اثر محافظتی آن پس از گذشت مدت زمان بیشتری از  RUUO بروز نماید، اما آسیب کوتاه مدت ناشی از UUO، می‌تواند حتی پس از RUUO  پیشرفت کند.     Background & aim: Unilateral ureteral obstruction (UUO) is a common disorder of the renal system that may cause irreversible kidney damage and depends on the duration of the obstruction and the extent of renal tissue damage and dysfunction. The aim of the present study was to determine the effect of estradiol on oxidative stress index and nitric oxide levels in unilateral ureteral obstruction in ovariectomized rats.   Methods: In the present experimental study conducted in 2017, 84 rats underwent ovariectomy (180 20 20 g) and were randomly divided into 14 groups of 6. Group 1 (sham group) had no surgery and all groups 2 to 14 had unilateral ureteral obstruction and some of them were treated with medication. Thus; Groups 2-4 were sacrificed 3 days after UUO and receiving the drug (sesame oil, 0.1 and 0.5 mg / kg estradiol), respectively, group 5-7 3 days after UUO and receiving the unblocked drug (RUUO) were sacrificed the next day, groups 8-9 and 10-12 were sacrificed 1 and 3 days after RUUO and estradiol, respectively, and groups 14-13 were sacrificed 3 days after UUO and RUUO and received the drug in both models. Were. Quantitative detection tests for stable metabolites of nitric oxide and malondialdehyde in blood and tissue samples were performed using special kits. The collected data were analyzed using statistical tests and LSD test.   Results: The results of the present study indicated that in the UUO model, the level of nitrite and MDA of renal tissue in the obstructive groups decreased significantly compared to the sham group (p <0.05). However, administration of different doses of estradiol did not alter serum and tissue levels of nitrite and MDA between the treatment and control groups in the UUO and RUUO models (p> 0.05).   Conclusion: According to the obtained results, it can be stated that estradiol administration during UUO and RUUO times had no effect on nitrite and MDA levels.
