fa تأثیر داروی لیراگلوتاید بر تغییرات بافتی رحم، هورمونهای استرادیول و FSH در موش نژاد NMRI Investigation of Liraglutide Effects on Changes of Uterine Tissue, Estradiol and FSH Hormones in NMRI Mice عمومى General پژوهشي Research چکیده زمینه و هدف: داروی لیراگلوتاید برای درمان چاقیهای مزمن و دیابت نوع دو، که از عوامل ناباروری می&lrm;باشند، مورد استفاده قرار میگیرد. با توجه به این که رحم به عنوان مهم‌ترین عضو دستگاه تولید مثل در معرض آسیب، به ویژه به وسیله داروهای شیمیایی میباشد، در این پژوهش تأثیر داروی لیراگلوتاید بر روی تغییرات بافتی رحم، هورمون‌های استرادیول و FSH در شرایط درون تنی بر روی موش نژاد NMRI بود.   روش بررسی:  این پژوهش تجربی، بر روی 28 سر موش ماده بالغ نژاد NMRI (8 تا 12 هفته‌ای) با وزن  بین 25 تا 30 گرم انجام گردید. موش‌های ماده به طور تصادفی در چهار گروه (هفت تایی) شامل؛ کنترل، شم و دو گروه تجربی تقسیم شدند. گروه کنترل بدون تزریق و به گروه‌های شم و تجربی 1 و 2 به ترتیب حلال دارو(آب مقطر) و لیراگلوتاید با دوزهای 6/0 میلی‌گرم بر کیلوگرم و 8/1 میلی‌گرم بر کیلوگرم تزریق گردید. تزریق به صورت روزانه و در یک ساعت معین به صورت زیرجلدی صورت گرفت.  پس از 30 روز تزریق، موشها عمیقاً به وسیله اتر بیهوش شده و ضمن خون‌گیری از قلب، رحم از بدن خارج گردید. به دنبال رنگ‌آمیزی، نمونهها با استفاده از میکروسکوپ نوری مورد بررسی بافتی قرار گرفتند. داده‌های جمع‌آوری شده با استفاده از آزمون آماری آنالیز واریانس یک طرفه (آنوا)  و تست توکی تجزیه و تحلیل شدند.   یافتهها: وزن موشها در گروههای تجربی 1و2 به طور معنیدار(01/0>p) و (001/0p<) در مقایسه با گروه کنترل و شم کاهش یافته است. هم‌چنین ضخامت جدار رحم (از لومن تا آخرین لایه) و نیز قطر لومن، به ترتیب کاهش معنی‌دار(05/0p<) و (01/0>p) را در گروه‌های تجربی نشان داد. ضخامت آندومتر در گروههای تجربی یک و دو به ترتیب کاهش معنی‌دار(01/0>p) و (001/0 p<) و هم‌چنین ضخامت  پریمتر در گروه تجربی دو کاهش معنیدار(05/0p<) در مقایسه با گروه کنترل و شم یافت. به علاوه در اثر استفاده از دارو، تعداد غدد رحم در گروه‌های تجربی یک و دو به ترتیب افزایش معنی‌دار(05/0p<) و(01/0>p) را نشان داد و نیز قطر آن‌ها در هر دو گروه افزایش یافت. هم‌چنین در میزان هورمونهای استرادیول و FSH به ترتیب کاهش و افزایش مشاهده گردید.   نتیجهگیری: درمان با لیراگلوتاید با کاهش وزن و ضخامت بافتی رحم همراه است. با توجه به اختلال در سیستم هورمونی، که احتمالاً به دلیل ساختار اینکرتینی دارو میباشد، هم‌چنین با توجه به کاهش هورمون استرادیول که هورمونی زنانه است، در اثر مصرف لیراگلوتاید سن باروری محدود خواهد شد.      Abstract   Background & aim: Liraglutide has used for treatment of obesity and type 2 diabetes, which are main factors of infertility. Regarding that, uterus as the most important organ reproductive system is under vulnerable, particularly by chemical drugs, the purpose of  this research was to investigated the effects of Liraglutide on uterine tissue changes and Estradiol and FSH hormones in NMRI mice in In vivo condition has been investigated.   Methods: This experimental study was performed on 28 adult NMRI female rats (8 to 12 weeks old) weighing between 25 and 30 grams. Mice were randomly divided into four groups (seven) including control, sham and two experimental groups. The control group was no injected and the sham and experimental groups 1 and 2 respectively received a solvent (distilled water) and liraglothedi at doses of 0.6 mg / kg and 1.8 mg / kg respectively. The injections were subcutaneously administered daily and in one hour. After 30 days of injection, the mice were deeply anesthetized by the ether and blood was removed from heart. After staining, samples were examined by optical microscopy. The collected data were analyzed using one-way ANOVA and Tukey's test.   Results: The weight of the mice in the experimental groups 1 and 2 significantly (p <0.01) and        (p <0.01) compared with control and sham groups has decreased. Also, the thickness of the uterus (from the lumen to the last layer) and the diameter of the lumen showed a significant decrease (p <0.05) and (p <0.01) in the experimental groups. A significantly decreased was seen on endometrial thickness in the experimental groups 1 and 2 (p <0.01) and (p <0.001). Also, the thickness of primer in the experimental group also decreased significantly (p <0.05) in comparison with the control group. In addition, using the drug the number of uterine glands in experimental groups 1 and 2 showed a significant increase (p <0.05) and (p <0.01), and their diameter increased in both groups. Also, estradiol and FSH levels were decreased and increased, respectively.   Conclusion: Treatment with liraglutadiene is associated with weight loss and tissue thickness of the uterus.
