fa شناسایی ساب تایپ‌های بلاستوسیستیس نمونه‌های انسانی با استفاده از ژن 18s rRNA در شمال غرب ایران Identification of Blastocystis sp. subtypes from human using 18s rRNA in Northwest of Iran انگل شناسی Parasitology پژوهشي Research چکیده زمینه و هدف: بلاستوسیستیس تک یاخته‌‌ای بی‌هوازی است که در دستگاه گوارش انسان زندگی کرده و می‌تواند به عنوان یک انگل زئونتیک در میزبان‌های مختلفی نیز وجود داشته باشد. این مطالعه در جهت شناسایی ساب تایپ‌ها و بررسی تنوع ژنتیک بلاستوسیستیس از نمونه‌های انسانی شهرستانهای ارومیه، تبریز و مراغه در شمال غرب ایران انجام گرفته است.   روش بررسی: در این مطالعه که به روش توصیفی ـ مقطعی است، 300 نمونه مدفوع انسانی که از  بهمن ماه 1395 تا مهرماه 1396  به صورت تصادفی و جهت غربالگری به مراکز بهداشتی و درمانی مراجعه کرده بودند، انتخاب و پس از مطالعه مستقیم میکروسکوپی مدفوع، 16 مورد ایزوله به دست آمده که  با روش DNA barcoding بررسی شدند. براساس روش‌های بیوانفورماتیکی سکانس محصولات به دست آمده مورد تحلیل و مقایسه قرار گرفتند.   یافتهها: از 300 نمونه مورد بررسی این مطالعه، تعداد 22 مورد به صورت مطالعه میکروسکوپی آلوده به بلاستوسیستس تشخیص داده شدند. 16 ایزوله متفاوت از موارد مثبت با روش مولکولی(PCR) جدا و سکانس محصولات به دست آمده مورد بررسی قرار گرفتند. 3 نوع ساب تایپ شامل؛ ST1، ST2 و ST3 در سکانس حاصل از این نمونه‌ها به دست آمد. یکی از نمونه‌های مورد بررسی پس از دوبار سکانس متفاوت، هر دو زیرگونه ST1 و ST3 را داشت، ولی با سکانس مجدد زیرگونه غالب فرد، ST3 گزارش شد.   نتیجه‌گیری: ساب تایپ‌های مختلفی از این انگل در این منطقه وجود دارد. با توجه به ماهیت ساب تایپها، چرخه زئونوتیک این انگل در منطقه وجود داشته و با شناسایی و تعیین زیرگونه‌های بلاستوسیستیس در میزبانهای مختلف به عنوان ارگانیسمی‌‌زئونوتیک، میتوان به نحوه انتقال انگل و مهاجرت‌های ژنتیکی آن دست یافت. به نظر می‌رسد میتوان با استفاده از الگوی ساب تایپ‌های به دست آمده انگل، در مطالعات‌ آینده، ارتباط آن با علایم بالینی مطرح و مورد بررسی قرار گیرد.     Abstract   Background & aim: Blastocystis sp. is an anaerobic parasite which lives in the digestive system and can be a zoonotic parasite in different hosts. This study was carried out to identify subtypes and to investigate the genetic variation of Blastocystis sp. from human samples in Urmia, Tabriz & Maragheh cities Northwest of Iran.   Methods: In the present descriptive-cross sectional study, 300 human stools from January to October 2017 were randomly selected from treatment centers in North West of Iran. Based on microscopic observation, positive samples examined by DNA barcoding methods for detection of Blastocystis sp. subtypes and their sequences were analyzed.    Results: of the 300 samples, 22 specimens diagnosed as Blastocystis sp. with the microscopic method. Sixteen apart isolates from the positive cases obtained by the molecular method (PCR) and the sequences of the products were examined. Three types of subtypes including ST1, ST2 and ST3 obtained from these samples. One of the samples after two different sequencings, was reported as both of the ST1 and ST3 subspecies, but re-sequencing of the dominant show ST3.   Conclusion: There are various subtypes of this parasite in this area. Due to the nature of the subtypes, the zoonotic cycle of this parasite exists in this region. By identifying and determining the Blastocysts subspecies in different hosts as a zoonotic organism, it is possible to detect genetic migration and parasite transmission.
