fa تعیین ارتباط پلی‏مورفیسم D/I ژن آنزیم مبدل آنژیوتانسین (ACE) با سقط مکرر جنین تخصصي Special پژوهشي Research زمینه و هدف: منظور از سقط تکراری وقوع 2 یا 3 بار سقط متوالی خود به خودی، قبل از هفته 20 می‌باشد. علل متعدی در ایجاد سقط تکراری در زنان نقش آفرینی می کنند که از آن جمله می توان به مشکلات انعقادی اشاره نمود. هدف این مطالعه تعیین ارتباط پلی‌مورفیسم D/I ژن آنزیم مبدل آنژیوتانسین با سقط مکرر جنین بود. روش بررسی: در این مطالعه مورد ـ شاهدی، 100 بیمار با سابقه سقط(حداقل دو بار) به عنوان گروه مورد و 100 خانم سالم بدون سابقه سقط به عنوان گروه شاهد انتخاب شدند. برای بررسی پلی مورفیسم‌های ژن آنزیم مبدل آتزیوتانسین(D/I) واکنش زنجیره‌ای پلی‌مراز با جفت پرایمر طراحی شده انجام شد. داده‌ها با آزمون آماری مجذور کای تجزیه و تحلیل شدند. یافته‌ها: ژنوتیپ I/I در افراد گروه مورد دارای فراوانی 7 درصد بود، در حالی که در گروه شاهد هیچ موردی مشاهده نشد(05/0p<). هم‌چنین فراوانی آلل I در گروه مورد و شاهد به ترتیب 6/36 و 24 درصد بود(05/0p<). نتیجه‌گیری: این مطالعه نشان داد، ارتباطی بین ژنوتیپ D/D و آلل D با سقط مکرر جنین وجود ندارد. Background & aim: Recurrent pregnancy loss (RPL) refers to the occurrence of two or more consecutive losses of clinically recognized pregnancies prior to the 20th week of gestation. The aim of this study was to investigate the hypothesis that recurrent pregnancy loss (RPL) is associated with a common insertion-deletion polymorphism in the angiotensin-converting enzyme gene.. Methods: The present paper intended to study the ACE deletion (D)/insertion (I) polymorphism in women with recurrent abortion. One hundred patients with recurrent abortions (at least two) as cases and one hundred healthy female with two or more normal term deliveries and without a history of abortion selected as controls. Genomic DNA was isolated from peripheral blood leukocytes and the ACE insertion/deletion (I/D) polymorphism in intron 16 was performed by polymerase chain reaction (PCR). For the statistical analysis, SPSS software version18 was used and Chi-square tests were calculated. Results: Seven patients, (7 %), and non from the control group, were homozygote (I/I) for ACE polymorphism (OR=22.82 95% CI=1.26-412.69 p=0.034), depicting no significant associations between ACE D allele or DD genotype and RPL.
