Antiprolifration Effect of Silver Nanoparticles Syntethised Using Centlla Asiatica on Breast Cancer Cell Line

Abstract:
Background and aim: The use of plants to synthesize nanoparticles is one of the most environmentally friendly methods. The purpose of this study was to synthesize green silver nanoparticles using Centella asiatica extract and study the effects of cytotoxicity in breast cancer cell line.

Methods: In this experimental study, the morphology of silver nanoparticles was evaluated using transient electron microscopy (TEM) and scanning electron microscopy (SEM). MTT was used to evaluate the anti-proliferative effects of biological nanoparticles in breast cancer cell line (MCF-7) and normal cells (HEK-293). Apoptotic assays were performed using flowchitometry with Annexin V kits. Data were analyzed using two-way ANOVA statistical tests.

Results: Spherical particles were synthesized with an average of 19.19 nm. Antimicrobial activity in MCF-7 cells increased with increasing concentrations and time of nanoparticles. IC50 of silver nanoparticle was 24.88 μg / ml in cancerous cells. Increasing apoptosis was observed in cells treated with biological nanoparticles at IC50 concentration compared to control cells (p <0.001). Also induction of apoptosis in MCF-7 cells was more than necrotizing compared to control (p<0.001).

Conclusion: The synthesized silver nanoparticles have the effect of cellular toxicity and induction of apoptosis on cancer cells in a concentration-time pattern.
